Reimu's job is to kill the Yokai, the japanese demons who threaten the earth. Marisa is the alternate playable character for more expert players. Over the course of the games, there's a variety of different threats to the earth. Whether it's the vampire mistress Remilia, or the black crow of hell, or rabbits from space. Throughout the games there's also a variety of other playable characters who are introduced. Usually popular characters from previous games. Like Reisen, or Sanae.

The lore is really silly. Apparently the girls all have unwritten rules and hold back on their attacks so they aren't unbeatable. With the exception of Junko, who is genuinely trying to kill you

According to Zun the Touhou universe has just as many males as females. But the guys "don't care for danmakou" so they don't engage in any of the battles.
