No it wasn't. You can play it keyboard only (and the base game is mostly easy enough that this is viable), but it was designed with using a keyboard and mouse in mind, you can see this in the included demo recordings, and there's video footage of John Romero testing the game during development using a mouse for turning, further even the Wolfenstein 3D manual suggested you use a mouse for turning.

The idea that Doom was designed for keyboard only comes from the fact that people with weak 386 computers didn't have enough memory to run both Doom and their mouse drivers at the same time.
Someone with a good 486 computer, with memory to spare, could enjoy the game As The Developers Intended (TM), and you will find that there's settings for a mouse in all versions of Doom on home computers.

Unless you're playing to challenge yourself, or you prefer a gamepad or joystick, there's no good reason to not use a mouse when you play.
