I think estimates are that about 5% of methane are unaccounted for. Also, some byproducts are assumed to come from natural sources because we don't seem to know otherwise. A lot of microbes seem to metabolize very slowly, though. Which is why it is not possible to cultivate them - the usual method to find out what they need to live and what chemicals they produce.

Explain the process needed. As far as I know we still use petri dishes and PCRs for detecting life which usually just works on regular life but not a different form of microbes.
