Not all writing is autobiographical. There's no definitive answer to the meaning of the metamorphosis, that's what makes it such a potent work of art.

But one thing you could submit is that it's about the dehumanization that people suffer under industrial society/modernity in general. People become streamlined and simplified (get to work on time, do tedious drudgery, go home, repeat) until they have become the simplest living thing possible: an insect. Insects straddle the line between living and unliving in an uncanny way; you could say that Kafka's genius was connecting this uncanny feeling to modernity as a whole.

There are many other interpretations you could offer, however. Personally I think Kafka is an underappreciated genius, but I dock him points for the sin of despair...

You do know that Kafka was highly unknown during the years he wrote and was writing the most of his stuff only for himself? He even told his bff Max Brod that he should burn everything written by him.
