I am pretty sure it's something picked up from otaku related works, probably from female artists referring to themselves through female characters. There was a concept mentioned in anime about people getting old and being single although usually it's more like 30, or has been since the mid 2000s. It's probably only ever been said in a couple things and probably also has ties to the same people who would say that 20 is a granny and 30 is a mummy. I know I've seen it at least once, though I can't recall what, as it was probably like 10+ years ago.

But I am 100% sure it's wholly something people picked up from anime and manga.

There is no air-tight argument as these are just emotionally-driven preferences. I never understood why people get attacked simply preferring one thing over another. Looking for a virgin is just eliminating that risk by not allowing you or your partner to compare your sexual ability with that of anybody else. There's nothing necessarily WRONG with not wanting to marry anyone who isn't a virgin, it just has no practical benefit so it's kind of stupid. The first time you have sex really isn't that big of a deal, it's just two animals smacking their bits together (usually very awkwardly, if it's their first time). If you take the proper precautions and don't get each other sick and/or pregnant then there's really no issue with having sex. That being said I don't think it's a great thing to constantly sleep around either, but if you have someone you really like there's no good reason to hold off until you get married. The practical benefit is that the two of you are less likely to suffer from sexual dissatisfaction, a leading cause of divorce and infidelity, because your experience is limited to each other. There have been plenty of studies that show a positive relationship between number of previous sexual partners and divorce rates. Strategic willful ignorance of certain intimate activities allows one to better focus on the more important aspects of a relationship that can't be improved so easily as sexual dissatisfaction can between people who haven't known anybody else. That's the way I see it at least. Others are free to slut it up if they want. I'll only object if they tell me that I should be doing the same.

Really though what I'm trying to say is the act of sex itself is fairly meaningless past what meaning we attribute to it (assuming you avoid pregnancy/spreading diseases). Someone having sex doesn't magically make them less able to have a happy marriage, in fact I'd say it lets them get in some practice beforehand. It might be the case that people who have a lot of sexual partners tend to be worse at commitment but in that case the person's mindset/personality is what's wrong, them having casual sex is just a result of that. Not everyone who has sex outside of a lifelong partnership does so because they suck at committing. It's not like you can ever really know someone will be your lifelong partner in the first place, especially with divorce rates so high nowadays.

I do think what you say about choice is correct though, I've seen studies showing that arranged marriages are actually generally happier and more successful than romantic ones.
