As soon as Irie came onto the scene, he processed incredible amounts of historical data in a short time, well beyond what anyone else would be able to achieve, forming a foundation the rest of the research was built upon. His efforts towards creating a cure would also require an exceptionally strong understanding of the mechanics of how the pathogen operates; Takano's research into H170 would certainly be based on that general research first and foremost. Irie simply had no interest in the subsequent work to produce a substance that would agitate the pathogen further, since that was of no benefit to anyone.

Takano was not at all prepared for Koizumi's death. She genuinely believed that the Tokyo higher-ups sincerely cared about the research, and that the research would still be able to continue without him, if obviously to a lesser extent, and was devastated to find out that Koizumi was the sole reason anyone ever supported the research, with nobody giving a single shit about Hifumi. For all of her posturing about how she wouldn't let a dice roll control her, she could do nothing to avert this one, only managing to embarrass herself in front of the board of directors.

And in her inner monologues during Matsuri, we repeated see Takano present Miyoko as having "died" in the orphanage. Her subsequent obsession with Hifumi('s research) is rooted in a severe lack of self-esteem, instead focusing her identity and reason for living entirely on her grandfather, which is absolutely not what he ever would've wanted. She threw away her humanity at that point and committed numerous heinous acts in Hifumi's name, when all he really wanted was for her to find her own happiness. Everything after that was just filler.
