I'd say it's hard to define their relationship with them either being friends or enemies. They were technically both.

As I've understood it, Japanese royal clan relations were complicated. The families served the Royal family all together and they all inter-married a lot, meaning all were related one way or another. But at the same time, there were family disputes, rivalries over different things, and the whole religious war.

Assuming they were both of high status among their clans in life, they probably grew up together and served the royal family side by side their entire life. But they would still be expected the see the other as an enemy depending on the circumstances.

That's how I've understood it. Someone correct me if I'm wrong.

As for how Futo tricked Tojiko into actually killing herself, she doesn't seem to have minded.
