With black pen:
You usually achieve depth by cross hatching, and varying thickness of the pen. Unless you're working with a nib for the drawing, where you can vary thickness by simply applying pressure on the nib, you'll need to reinforce certain lines and make them thicker - and some thinner (by inclining the pen to "scratch" the paper, or using thinner/thicker pens). follow artist who do this a lot - examples that come to mind are takehiko inoue and kentaru miura, but just Google cross hatching faces. And notice what they do, copy it, repeat. I really think the best way to get this is practice and observation.

Remember with faces, with pen LESS IS MORE. Don't make the mistake of putting too many lines, you NEED white, or the drawing will look like a overdone mess..much of ink is knowing when to stop.

With hair, and this applies to both pencils and pen, think of it as volume, not individual strands of hair. It's clumps youre drawing, so try to get that the direction and feel, then fill it up (dark parts first imo), and always, always, colour/shade in the direction the hair is going. Finish individual hairs at the end with the pen/pencil for a more realistic look.
