When a French word (in Canadian French pronunciation) ends in vowel-N-E, then the N is distinctly pronounced, and the preceding vowel has a long pronunciation. If a word ends in vowel-N, then the N becomes an unpronounced nasal voiced end, and the vowel has a short pronunciation. "Poutine" has a distinct "een" ending, whereas a word like "poutin" would have more of an "enh" ending.
Furthermore, P-O-U as the start of a word is not completely pronounced like "poo", but it's not even close to "pu" (as in "put"), and compared to "pu" is actually much closer in pronunciation to "poo".
The whole "ts" or "tz" part is just French-Canadians being insufferable hipsters who are too "cool" and "aloof" to pronounce things clearly, instead of slurring everything together in some attempt to obscure their language from all those "outsiders" that keep somehow easily learning it.

So yes, you fuck, it is almost exactly pronounced "POO-TEEN", except the emphasis is put on the final syllable, so the "OO" sound in the "POO" part isn't enunciated quite as hard, thus, it's more like "pooTEEN".

In practice, the I is rounded considerably, but not made into a short I (it's not Putin). It's almost like it should have a circonflexe, but they remembered that it doesn't really have an effect on I. If poutisinal was a word, more English speaking people would get that I right.
