I don't know which wallets use 2fa.
I bought a Ledger when I first started using crypto so I'm not very knowledgeable in the alternatives.
But if you're suggesting keeping your tokens in an exchange account, I'll have to defer to the "not your keys, not your coins" trope. Which goes back to my "personally devastating" remark. I'm uncomfortable handing my money to a third party and hoping they'll give it back when I ask for it.

its never safe to leave them in an exchange. 2fa protects access to the web interface to the exchange, but not the actual keys themselves.

My portfolio is barely crossing 10k right now and I have been thinking about it... However, I have always seen these things as glorified encrypted USB drives. Ledger is even full propietary stuff.

I spread my crypto into 2 cold wallets made in an airgapped PC, and for hot wallets I have used Trust Wallet and MEW. The backups and seeds I have them encrypted on Veracrypt volumes. I have had zero issues these past two years... I am pretty confident in my security approach but I won't say these things can't be useful.

I'm at about 100k right now and it's split between a cold wallet made on an airgapped PC and my coinbase account. I'm at the point where I need to start moving some of the coinbase btc over to the cold wallet again. I keep some in there in case I need cash fast, but since I also keep cash in a lockbox it's probably not necessary.

I guess if I ever need to start spending BTC regularly I'll get a hardware wallet and transfer like half a BTC over to it, but for the most part I think paperwallet is safest.
