Okay so with pk issues, the slide that CRNX showed (pic related), it does not make sense to me what dosing up to 60 is supposed to do when 20/30 is pretty much flatlining. They have a new formulation but the exposure isn’t really that much better, they have lower cmax/AUC at 20/30mg comparatively which then show a greater dose response at higher doses, but the AUC is in line with the old formulation.

Another skeleton in CRNX's closet is that they never showed us the results based on the Phase 3 endpoint responder analysis which is IGF-1 <1.0 ULN…they only showed mean change of IGF-1. Which is a really bad measure esp when we know this drug suppressed IGF-1 substantially in some patients.

As to the 60mg dosing I can only assume, as you are doing without any evidence, that they are investigating the upper dose, I agree it seems like a moot point given the decreased effectiveness over the 20-30mg range but we don't have access to ongoing research data. Finally they report the mean change of IGF-1 because the trial is comparing the oral drug with the IV drug, attempting to demonstrate equal effectivness with limited side effects.
Will this drug be wildly successful, I doubt it in the long run due to the limited population size with this disease pathology. But to short the stock over a binary event I think is a mistake. I sincerely doubt the P3 trials will be that disappointing. It seems you're basing your analysis on a lot of misguided presumptions without a strong background in understanding the nitty gritty of clinical trials. If you feel its a good short option, go for it.

I still think their worse AEs leads to underpowering issues. They're powering for 20% placebo and 70% active arm responder rates.

Look more into CRNX data and their competitor, Mycappsa, and another R&D phase acromegaly drug from the Swedish company Camarus. A couple weeks ago Camarus reported 38% pbo response at week 24. Bodes poorly for CRNX.

Mycapssa is the standard of care for oral and Mycappsa's 58% response rate was viewed as a disappointment, though it was consistent with other acromegaly pivotal studies

Even though Mycapssa has consistently shown an ability to maintain IGF-1 levels >1x ULN, patients often saw an absolute increase in their IGF-1 levels while on Mycapssa in both OPTIMAL and MPOWERED versus the level maintained on injectables.

I would assume pic related is why they upped dosing. Is odd that it doesn't align w/ pk.

I think the Phase 3 trial actually hits but the stock still goes down. I think acrogelamy is a small market anyway and my base case is trial hits but just not differentiated from mycapsa, which imo sends it down to low mid teens. That is why CRNX is a short.

It's not that the Phase 3 trials miss their endpoint cleanly, but that the data isn't good enough for the market to warrant it a commercial success in a competitive acromegaly market.

If you look at the majority of clinical trials for any drug of any class the trials typically present the same. If you've been following clinical drug trials for any amount of time you'd catch on that most drugs have minimal effectiveness in vivo compared to competitors. There's no real innovation or novel drugs being developed. Yet the companies will pump or get bought out on hopium. So it seems like we agree on the likely path of this trial I disagree with your conclusion that that company stock is going to drop. Back test a wide variety of drugs and the stock changes and you'll find there is not a lot of correlation between the actual efficacy of a drug and the stock price. Also the pic you posted is demonstrating that igf-1 levels remain low even after discontinuation of the drug. This is different information than the other pics you've been posting. And symptoms aren't marked as treatment related because they are patient reported symptoms. There is no proof one way or the other that they are related to the drug. This is standard in trials. A person can be enrolled in a trial and have a headache that is because of poor sleep. But if they report it to the investigator it gets reported as declared symptoms. I don't know what your background is but there is a lot more to clinical research trials than numbers and I would suggest you read up on more trials to get a feel for how they are formated.

There is no system, you have to develop your intuition to where you can tell if traders are getting fucked or not and avoid traps. The setup or signal could be something very simple like
>price rejecting down from a lower high, wedge bottom, any other price action pattern
>shows of force like engulfing candlesticks, big wicks, big ass bars closing on their low or high, consecutive bull or bear closes above or below a moving average
>moving average crossovers, RSI spiking hard off support or resistance, or whatever indicator you want to use
Basically just define some clear setup and watch the market for opportunities, then git gud at reading the situation correctly and catching moves but also not getting trapped on fakeouts. It's not the signals or patterns that are most important, they just serve as a means to clearly present your brain with a situation. The magic is in how you interpret things. Over the years you may get better and better at playing your cards right, just keep pushing and fighting in the trenches until you get where you need to be.
And obviously control risk and your emotions, use a small position size until you're more consistent. If you can a wide stop is helpful combined with a small position size so you can still have low risk. If not at the very least have it above or below the relevant major support or resistance so you have a fighting chance. Eventually you may be good enough to use tighter stops.

I was shorting the Nasdaq using futures since January 25th. As you can fucking imagine, that hasn't worked out too well. After losing 50% of my account, I (finally) capitulated on Friday after yet another FUCKING gap up and return to the June 16th highs.

But now, this whole week, it's just frozen at Friday's levels, and futures look pretty red tonight.

For fucking God's sake, please do not tell me that I held short through five months of pure agony and capitulated at the exact top. Tell me that the market is going to go up another 15% from here.

I'm not sure what it is called. In the same way you see seasons when tracking the sun through the zodiac, you see market seasons when tracking planets.
Saturns cycle is 30 years or so with booming markets from Libra until Aries. Markets are less successful from Aries to Libra.
2025 is less growth.
2010-2025 boom
1995-2010 crabby
1980-1995 boom etc.
Of course, this isn't the only force on the markets, it's just what Saturn does.
Mars is the opposite and only takes two years or so for a cycle. From Aries to Libra is good, Libra to Aries is less so.
Jupiter is weird. Its benificial all the time except from middle of Scorpio to middle of Aquarius.
