If it closed at 321, you'd need the requisite funds to purchase 100 shares of SPY at the strike of 320, and if you did, then it'd execute for you. At that point you could sell the SPY you bought at 320 for 321 and make your profit

If you dont have the money to buy the 100 shares at the strike price, Robinhood wont execute even if the call is in the money, so you'd want to sell back the call before its expiry
